L-DOPA BIOSYNTHESIS WITH Agaricus bisporus TYROSINASES ASSISTANCE

نویسندگان

چکیده

L-DOPA (3,4-dihydroxyphenyl-L-alanine) is a drug of choice in Parkinson's disease treatment. However the chemical method its synthesis has number drawbacks, so biotechnological approaches are being explored as an alternative. Aim. The goal to develop new affordable and effective biosynthesis using mushroom tyrosinase, immobilized economical carrier, which ensures stability enzyme multiple uses. Methods. Agaricus bisporus isolated tyrosinase was used work. carried out aqueous organic medium. obtained product analyzed mass spectrometry, specific rotation melting point. immobilization poly-N-vinylpyrrolidone (PVP), interaction with pH-optimum application frequency were determined. Results. A partially purified preparation from bisporus. In solution presence, only 5.1% due subsequent formation complex polycyclic compounds. derivative methylene chloride addition buffer made it possible obtain yield 55%. Tyrosinase PVP showed activity 30% higher than free CH2Cl2 medium biocatalysis for 7 cycles. Conclusions. synthesizing available biocatalyst based on developed, enabled during cycles use

Mushrooms (Agaricus bisporus) mediated biosynthesis of sliver nanoparticles, characterization and their antimicrobial activity

In this paper we report an eco-friendly route for the synthesis of sliver nanoparticles using Agaricus bisporus (white button mushroom) extract. The synthesized silver nanoparticles were confirmed and characterized by UV-Visible spectrum of the aqueous solution containing silver ions showed a peak at 420 nm corresponding to the surface plasmon absorbance of silver nanoparticles. Transm...

Agrobacterium tumefaciens mediated transformation of Agaricus bisporus

Agrobacterium tumefaciens is known to transfer parts of its tumor-inducing plasmid to filamentous fungi including Agaricus bisporus. We have used this transformation system to transform germinating basidiospores of A. bisporus. Analysis of hygromycin B-resistant clones showed that the T-DNA integrated at random sites and in multiple copies into the host genome. Furthermore we have been able to ...

Virus-Enriched Fraction from Agaricus bisporus

Partially purified virus preparations from sporophores of Agaricus bisporus affected with LaFrance disease had up to a 15-fold-higher RNA-dependent RNA polymerase activity than did comparable preparations from healthy sporophores. Enzyme activity was dependent upon the presence of Mg2+ and the four nucleoside triphosphates and was insensitive to actinomycin D, a-amanitin, and rifampin. The 3H-l...

Using tea waste as a new casing material in mushroom (Agaricus bisporus (L.) Sing.) cultivation.

In this study, the possibility of using tea production waste as a new casing material in mushroom (Agaricus bisporus) cultivation was investigated. Some physical and chemical characteristics of tea waste, fermented tea waste and a mixture of tea waste with peat were compared with that of peat casing, as were their effects on yield. The highest yield was obtained from peat casing. Using tea prod...
